To Raise Cain means to cause trouble and uproar, an American expression that dates from the early 19th century. ‘Raise’ Cain here is used in the sense of ‘conjuring up’ Cain. Cain was cursed by God in the Bible for murdering his brother Abel and thus, when one conjures up Cain, one is conjuring up trouble.
